Peripheral blood cultured mast cells: Phenotypic and functional outcomes of different culture protocols

چکیده

Mast cells (MCs) play a pivotal role in innate and adaptive immune responses. However, MCs are also involved different pathologic conditions. Studies on the mechanisms that govern human MC functions impeded by their limited difficult recovery. Therefore, several research groups have developed protocols to culture from progenitor cells. These vary with respect duration used maturation cytokines. How obtained differ phenotype functionality is currently unknown. To compare for generation of peripheral blood progenitors. Thirteen paired cultures were investigated. cultured form CD34+ progenitors 4 or 8 weeks without addition IL-6. Phenotyping comprised staining CD117, CD203c, Fc?RI, MRGPRX2, CD300a CD32. Functional studies included measurements up-regulation CD63 CD203c after allergen-specific cross-linking sIgE/Fc?RI complexes ligation MRGPRX2 substance P drugs. Cell presence IL-6 consistently yielded highest numbers MCs. showed more autofluorescence significantly impeding correct analyses Fc?RI The density CD32 was comparable between expression higher week cultures. increased Cells responsive activation. In contrast, 4-weeks Four sufficient generate sizeable mast progenitors, thereby enabling simultaneous exploration non-specific activation via MRGPRX2.

Conditioned Medium from Cultured Colorectal Cancer Cells Affects Peripheral Blood Mononuclear Cells Inflammatory Phenotype in Vitro

Background: Colorectal cancer (CRC) is the third most common cancer worldwide. Studies have indicated that immune cells and soluble factors play a key role in maintaining the balance between tumor-promoting inflammation and anti-tumor immunity. It has been shown that secreted cytokines from CRC cell lines could affect peripheral blood mononuclear cells (PBMCs), monocytes, and macrophages phenot...

Culture and Expansion of Rat Mesenchymal Stem Cells Using the Serum Prepared From Rat’S Peripheral Blood

Purpose: The aim of this study is to evaluate the effects of prepared serum from rat peripheral blood on growth and viability of rat bone marrow-derived mesenchymal stem cells (MSCs) in vitro. Materials and Methods: Bone marrow cells prepared from rat’s tibia and femur central canal were cultured either in medium containing fetal bovin serum (FBS) or rat peripheral blood-derived serum (PBDS) f...

Comparison of Functional Competence of Umbilical Cord and Adult Peripheral Blood Dendritic Cells in Allogenic Mixed Leukocyte Reaction

Background: Dendritic cells (DCs) are the most potent stimulators of primary T cell responses and play a key role in immune reactions after stem cell transplantation. Very little is known about the cord blood (CB) dendritic cells and their potential involvement in the low incidence and lower severity of acute graft-versus-host disease after CB transplantation.   Objectives: The aim of this stud...

Phenotypic and Functional Characteristics of Blood Natural Killer Cells from Melanoma Patients at Different Clinical Stages

Melanomas are aggressive skin tumors characterized by high metastatic potential. Immunotherapy is a valuable alternative for metastatic melanoma patients resistant to chemotherapy. Natural Killer (NK) cells are efficient anti-tumor cytotoxic effectors. We previously showed that blood NK cells from stage IV metastatic melanoma patients display decreased NK receptors and that chemotherapy modifie...

In Vivo Effects of Calcitriol on Phenotypic and Functional Properties of Dendritic Cells

Background: Dendritic cells (DCs) play a central role in the initiation and expansion of T cell mediated immune responses with potential immunotherapy application. The compounds which have the ability to induce immunomodulatory effects on DCs may be employed for the treatment of immunopathologic conditions such as autoimmune diseases. Objective: The aim of this study was to investigate the in v...
